Debutà al cinema el 1952 en Don't Bother to Knock , de R.W. Baker i el 1962 aconseguí l’Oscar a la millor interpretació per The Miracle Worker d’A. Penn, paper que ja havia representat en el teatre. Participà, entre d’altres, en els films The Pumpkin Euter (1964) de J. Clayton, The Graduate (1967) de M. Nichols, The Turning point (1977) de H. Ross, Agnes of God (1986) de N. Jewison, Torch Song Trilogy (1988) de P. Bogart, G.I.Jane (1997) de R. Scott, Critical Care (1997) de S. Lumet, Great Expectations (1998) d’A. Cuarón, Keeping the Faith (2000) d’E. Norton, i Up at the Villa (2000), de Ph. Haas. El 1980 dirigí Fatso . Estigué casada amb el director Mel Brooks.
