Fill del poeta Cecil Day-Lewis i de l’actriu Jill Balcon. Després de protagonitzar The Unbearable Lightness of Being (P. Kaufmann, 1987), obtingué el BAFTA i l’Oscar del 1989 al millor actor per My Left Foot, de J. Sheridan, pel·lícula que el confirmà com un excel·lent actor dramàtic. Altres films seus són Gandhi (R. Attenborough, 1982), Stars and Bars (P. O'Connor, 1988), The Last of Mohicans (M.Mann, 1992), The Age of Innocence (M. Scorsese, 1993), In the Name of the Father (J. Sheridan, 1993), The crucible (N. Hytner, 1996), The Boxer (J. Sheridan, 1997), Gangs of New York (M. Scorsese, 2002) pel qual aconseguí el premi BAFTA com a millor actor, i The Ballad of Jack and Rose (R. Miller, 2005). L’any 2008 guanyà de nou els premis BAFTA i l’Oscar com a millor actor, i el Globus d’Or per la seva interpretació a There Will Be Blood (2007), de Paul Thomas Anderson. Després de Nine (2009), de Rob Marshall, amb Lincoln (2012), de Steven Spielberg, obtingué un tercer Òscar (l’únic actor en aconseguir-lo), a més de un BAFTA i un Globus d’Or.
