Estudià art dramàtic a Yale i s’incorporà a l’Actor's Studio de Nova York. Debutà com a actor teatral a Broadway el 1953, i l’any següent debutà en el cinema en The Silver Chalice , de V. Saville. Posteriorment actuà entre d’altres, en els films de R. Wise Somebody Up There Likes Me (1956) i Until they sail (1957), Cat on a Hot Tin Roof (1958, R. Brooks), The Long, Hot Summer (1958; M. Ritt, pel qual fou premiat a Canes), The Young Philadelphians (1958, V. Sherman), Exodus (1960, O. Preminger), The Hustler (1961, R. Rosen), Hud (1963, M. Ritt), Harper (1966, M. Ritt), Butch Cassidy and Sundance Kid (1969, G. R. Hill), The Macintosh Man (1973, J. Huston), The Sting (1973, G. R. Hill), The Towering Inferno (1974, J. Huillermin i I. Allen), Buffalo Bill and the Indians (1976) i Quintet (1979), de R. Altman, Absence of Malice (1981, S. Pollack), The Verdict (1982, S. Lumet), The Color of Money (1986, M. Scorsese), amb el qual obtingué l’Oscar d’interpretació masculina, Blaze (1988, R. Shelton), Fat Man and Little Boy (1989, R. Joffé), Mr. and Mrs. Bridge (1990, J. Ivory), The Hudsucker Proxy (1994, J. Coen), Nobody's Fool (1994) i Twilight (1998), de R. Benton, Message in a Bottle (1999, L. Mandoki), Where the Money Is (2000, M. Kanievska), Road to Perdition (2002, S. Mendes), després del qual es retirà, tot i que continuà participant en algunes produccions televisives. Esdevingut una estrella de Hollywood, el 1968 inicià la seva activitat com a realitzador amb Rachel, Rachel i continuà amb The Effect of Gamma Rays on Man in the Moon Marigolds (1973), l’autobiogràfica Harry and Son (1982) i The Glass Menagerie (1987). El 1994 guanyà un Oscar honorífic. Casat amb l’actriu Joanne Woodward, destacà sempre pel seu compromís actiu en polítiques d’esquerra i en activitats benèfiques. Per aquestes activitats, el 1994 guanyà un Premi Humanitari Jean Hersholt, atorgat també per l’Acadèmia dels EUA.
