S’inicià el 1980 en una sèrie per a la televisió. Debutà al cinema amb Splash! (R. Howard, 1984) i s’especialitzà en el gènere de la comèdia amb títols com: The Money Pit (R. Benjamin, 1986), The Burbs (J. Dante, 1988) i Big (P. Marshall, 1988). La seva versatilitat li ha permès també interpretar drames i obtenir els Oscar del 1993 i del 1994 per Philadelphia, de J. Demme, i Forrest Gump, de R. Zemeckis, respectivament.
Després interpretà The Bonfire of the Vanities (B. de Palma, 1990), Joe Versus the Volcano (J.P. Shanley, 1990), Sleepless in Seattle (N. Ephron, 1993), Apolo XIII (R. Howard, 1995), Saving Private Ryan (S. Spielberg, 1998), Cast Away (R. Zemeckis, 2000), Road to Perdition (S. Mendes, 2002), Catch Me If You Can (S. Spielberg, 2002), The Ladykillers (E. i J. Coen, 2004), The Terminal (S. Spielberg, 2004). També protanonitzà The Da Vinci Code (2006) i Angels & Demons (2009), de Ron Howard; Charlie Wilson's War (2007), de Mike Nichols; The Great Buck Howard (2008), de Sean McGingly; Extremely Loud & Incredibly Close (2011), de Stephen Daldry; Cloud Atlas (2012), de Tom Tykwer i Lana Wachowski; Captain Phillips (2013), de Paul Greengrass; Saving Mr. Banks (2013), de John Lee Hancock; Bridge of Spies (2015), de Steven Spielberg; Ithaca (2015), de Meg Ryan; A Hologram for the King (2016), de Tom Twyker; Sully (2016), de Clint Eastwood; Inferno (2016), de Ron Howard; The Circle (2017), de James Ponsoldt i The Post (2017), de Steven Spielberg.
És també productor de cinema i de televisió i h a dirigit alguns telefilms i, per al cinema, els llarmetratges That Thing You Do! (1996) i Larry Crowne (2011), que també interpretà.
