Després de dedicar-se a la música rock, debutà en el cinema amb la pel·lícula de terror A Nightmare on Elm Street (1984), de W. Craven, i obtingué popularitat gràcies a la sèrie de televisió 21 Jump Street. Cry Baby (1990), de J. Waters, i la pel·lícula Edward Scissorhands (1990), de T. Burton, que l’especialitzaren en personatges de característiques excèntriques, un tipus de paper que ha interpretat molt sovint. Ha intervingut a Arizona Dream (1993), d’E. Kusturica; Benny & Joon (1993), de J. Chechick; What’s Eating Gilbert Grape (1993), de L. Hallström; Ed Wood (1994), de T. Burton; Don Juan de Marco (1995), de J. Leven; Dead Man (1995), de J. També ha protagonitzat les sèries cinematogràfiques Pirates of the Caribbean —The Curse of the Black Pearl (2003), Dead Man’s Chest (2006), At World’s End (2007), On Stranger Tides (2011) i Dead Men Tell No Tales (2017)—, interpretant el personatge del capità Jack Sparrow, i Alice in Wonderland (2010) i Alice Through the Looking Glass (2016), com a Tarrant Hightopp (el Barreter). Ha dirigit i protagonitzat The Brave (1997).
L’any 2021 fou guardonat amb el premi Donostia del Festival de Cinema de Sant Sebastià.
